Peter would be born to two agents of S.H.I.E.L.D., Richard and Mary Parker, who worked directly under Nick Fury Jr. Peter would commonly be left to stay with his Uncle Ben and Aunt May while his parents were on missions. However, at age 4, Peter would be left at his relatives' house for what should have been a few months, but they never returned, with the only information being that their plane had crashed in France. His Aunt and Uncle would adopt him and raise him as their own.

Shortly after moving into Forest Hills, Queens, Peter would befriend Mary Jane Watson, whose family lived next door. Peter and Mary Jane, aka MJ, would find they have a deep bond that transcended time and space. Peter would meet Gwen Stacy while attending Midtown Elementary School, as well as Eugene "Flash" Thompson. Peter would meet a lost Harry Osborn in Queens after having snuck out of a science event Peter was trying to see from outside, leading Peter to bring Harry home, where they would create a lasting friendship. Lastly, Peter and Harry would be saved by bullies by Eddie Brock, who would act as the group's older brother figure.

The group would grow distant as they aged, but would begin to come back together during their time in high school.

At the age of 15 years old, Peter would visit a Horizon Labs science exhibition where he would be bitten by a spider (Parasteatoda Tepidariorum) that had been exposed to radioactive rays from the radioactive isotope experiment. This would grant him the amazing powers of a spider.

Peter would go on to use these newfound powers to gain money from local wrestling competitions while beginning to stand out at school. Peter would receive advice from his Uncle and Aunt as they noticed the changes in his personality and demeanor, which he would ignore at the time. Peter would continue to win money from wrestling, until one night when he was shorted on his pay, Peter would allow a robber to steal from the wrestling organizers, allowing him to escape without helping, brushing the issue off as not his problem.

Later that night, Peter would discover his Uncle Ben had been shot while looking for him. His uncle would die in his arms before giving chase to the gunman, who was currently being pursued by the police. Peter would watch the criminal in a warehouse, shocked by the discovery that it was John Doe, the very criminal he had allowed to escape earlier that night.

Ridden by guilt, Peter would dedicate his life to helping people and protecting life as the Amazing Spider-Man.

Peter would go on to marry and have children with Mary Jane Parker-Watson, Richard, and Mayday Parker.
