Jamestown was the first permanent settlement in the Americas. It was going to fail from the start. Many of its colonists weren't prepared to sustain the colony for a long period. They became focused on becoming rich and didn't focus on more important things such as gathering supplies and food. The consequences then came to haunt them as they did not last very long through the harsh winter months, diseases, and Indian attacks.
