Smithies was able to take cells from bladder tumors, and replace their copies of the beta-globin gene without using a vector. This was a surprising result since all he had done was combine the new DNA with calcium phosphate and submerge the tumor cells in the mixture. He was able to observe that the cells had engaged in HR and had replaced the old DNA with the new synthetic DNA (Doudna 24).
