Manifest Destiny was the belief that the expansion of the US throughout the American continents was not only justified, but inevitable. Manifest Destiny helped lead to the Civil War because it led to free-soilers demanding that the land remain slave-free. The South, on-the-other-hand, wanted to expand slavery. Eventually most of the territories in question became free which angered the South. (if the territories became free states this added anti-slavery votes in Congress which could lead to the end of slavery) This added tension between the North and South which in turn helps lead to the Civil War.
