After the Louisiana Purchase, there were major debates over the slavery status of the land they had gotten. Prior to the inclusion of Missouri, the nation had an equal split of slave and free states, which gave the nation at least some degree of equality in that area. The introduction of Missouri would tilt the scales in one direction or another, so a compromise had to be drawn up.
Missouri was admitted as a slave state, while Maine was separated from Massachusetts and made into a free state, essentially adding one slave and one free state to the nation. After that, every state above the 36 30 longitudinal line was considered free.
This was significant because it helped maintain a balance between slave and free states and added a separation line (image attached).
